Madekwe was born South London, England into a mixed family. She's English but her father is Nigerian. She was born on an estate in South London community estate, and when she was fourteen, she attended The Brit School. She earned her BA in acting from the Royal Academy of Dramatic Art. She performed in various stage productions during her school days, including the stage productions King Henry V and Wuthering Heights.

Madekwe made her US debut in the 2009 drama series The Beautiful Life, which only aired two episodes on The CW before being cancelled.
